内分泌干扰物与发育毒性研究组课题组简介
研究组主要基于模式生物开展内分泌干扰相关的发育毒性研究,同时结合典型污染现场的人群调查,探讨化学污染物对人体健康潜在的危害。
主要研究兴趣:
1.环境激素导致的两栖动物性逆转及分子机制的研究
在国际上较早使用生物学研究的模型动物非洲爪蟾开展生殖内分泌干扰研究,发现非洲爪蟾性腺分化发育和第二性特征发育对环境雌激素高度敏感,可被诱导发生由雄到雌的性逆转或部分的雌性化。首次证实我国本土两栖动物种黑斑蛙对环境雄激素敏感,可被诱导发生由雌到雄的性逆转或部分的雄性化。利用非洲爪蟾和黑斑蛙各自的特点,分别建立了研究环境物质雌激素效应和雄激素效应的动物模型。应用这两个模型,研究组广泛研究可疑环境性激素对两栖动物性别分化的影响,并积极探索环境激素通过核受体导致两栖动物性逆转的分子机制。实验室建条件成熟的非洲爪蟾养殖动物房和黑斑蛙养殖动物房,可面向国内科研单位提供实验动物。
2.化学污染物的甲状腺干扰作用及相关发育毒性的研究
两栖动物变态发育受甲状腺激素(TH)调控,TH受体TR介导的信号通路及其所调控的发育过程在所有脊椎动物具有高度保守性。利用两栖动物变态发育的特点,研究组在建立研究甲状腺干扰作用的非洲爪蟾模型的基础上,对新型污染物的甲状腺干扰作用及相关的发育毒性进行系统研究,尤其关注对脑发育的影响。同时,发展简单易行的毒性评价方法,以形成国家/行业标准,为我国化学品环境管理提供技术支持。
3.电子垃圾污染区生物富集及健康风险
在台州电子垃圾拆解区开展PBDEs、得克隆等新型污染物的生物富集、人体负荷/健康风险研究:证明水生、陆生和两栖动物富集PBDEs的规律存在差异;发现较为高等动物脑中PB
DEs含量远低于其它组织中含量,显示血脑屏障对PBDEs存在屏障作用,且对高溴代物的屏障作用高于对低溴代物的屏障作用;在人体胚胎中检出PBDEs,证明在发育早期PBDEs即可进入胚胎,对胚胎发育可能存在一定的健康风险;发现DP及其类似物可在生物体/人体中累积到较高的水平,母体中累积的DP及其类似物可进一步传递给子代,使子代处于产前/产后的双重暴露风险之中。
